A meme is a context-free unit, i.e. one that does not require explanation. Most often, the humor in memes is so simple and easy that everyone understands it.
Memes can be intentional, created by marketers or users in the hope of virality. Co-opted — those that arose spontaneously, were picked up by people and went viral. And also self-generated — created by the "residents" of the Internet themselves, which went viral (UGC — User Generated Content).
It is better to use a ready-made template and make adaptations from already viral memes than strained intentional ones. Because they can go unnoticed or, even worse, misunderstood.

Jokes: how to come up with them?
A joke usually consists of two parts: the setup, the first part that creates anticipation, and the punchline, the second part that is responsible for surprise and makes you laugh. The comic effect occurs when there is a point of change between these two parts - a word or action that can be interpreted in different ways: we understand it in one context, but the essence turns out to be in another. And it is this contradiction with the usual course of affairs that creates a joke.
